A fate map for the larval epidermis of Drosophila melanogaster: localized cuticle defects following irradiation of the blastoderm with an ultraviolet laser microbeam.

چکیده
Drosophila melanogaster embryos at the cellular blastoderm stage were irradiated with a uV laser microbeam (257 nm, 7 erg) of 20-q diameter. Depending on the site of irradiation, up to 90% of the resulting first-instar larvae showed defects in the integument. The location of the defects within the cuticular pattern corresponded closely to the site of irradiation. The data indicate that the anlagen for the left and right parts of the larval epidermis are separated ventrally and each occupies an area extending from 20 to 60% of the egg length (EL) in the anteroposterior direction (0% EL = posterior pole). Within this area, the anlagen for the three thoracic and eight abdominal segments are equally spaced, each occupying a transverse stripe of a width of about 3.9 EL, corresponding to three to four cell diameters. The region of the blastoderm from which the epidermis of these segments develops comprises about 2200 cells which is one-third of the cells of the blastoderm. The paper includes a detailed description of the cuticular pattern of a Drosophila melanogaster fust-instar larva.

Localized ultraviolet laser microbeam irradiation of early Drosophila embryos: fate maps based on location and frequency of adult defects.
Drosophila embryos were locally irradiated with a 257-nm laser microbeam during blastoderm and germ band stages. Depending on stage and beam diameter (IO-30 pm), from 0 to 45 nuclei were exposed to the uv radiation. The doses used, 5 or 10 erg, did not eliminate nuclei or cells at once, but up to 50% of the adult survivors from irradiated eggs carried defects in the thorax. These were scored wi...

Bombyx eggs at the fertilization stage (0-2 hours after oviposition) were irradiated with a scanning UV-laser microbeam (355 nm) over an area of about 1% of the total egg surface. In spite of absence of nuclei or cells at the irradiated sites, larvae from treated eggs showed localized cuticle defects in the integument. The location and frequency of the defects within the cuticular pattern corre...

Background: In this study, the toxicity of the different xenobiotics was tested on the fruit fly Drosophila melanogaster model system. Methods: Fly larvae were raised on food supplemented with xenobioticsat different concentrations (sodium nitroprusside (0.1-1.5 mM), S-nitrosoglutathione (0.5-4 mM), and potassium ferrocyanide (1 mM)). Emergence of flies, food intake by larvae, and pupation h...
متن کاملGenetic control of cuticle formation during embryonic development of Drosophila melanogaster.
The embryonic cuticle of Drosophila melanogaster is deposited by the epidermal epithelium during stage 16 of development. This tough, waterproof layer is essential for maintaining the structural integrity of the larval body. We have characterized mutations in a set of genes required for proper deposition and/or morphogenesis of the cuticle. Zygotic disruption of any one of these genes results i...
متن کاملFate map and cell lineage relationships of thoracic and abdominal mesodermal anlagen in Drosophila melanogaster
We have examined the cell lineage of larval and imaginal precursors of the mesodermal anlage between 10% and 60% egg length (EL) by homotopic single-cell transplantations at the blastoderm stage. Clones in the larval somatic muscles and in the fat body were derived from transplantations everywhere between 10% and 60% EL along the ventral side of the embryo. Clones frequently overlap these tissu...
